I have one simple test that I apply to all Web designs that I evaluate: Apple-+ (Apple+Plus).
Try it, and you'll notice where layouts break, graphics are used instead of text, space is limited, and a host of other basic problems. Flexibility is a harsh mistress, but it is also the mark of simplicity.
Two great Web apps that stand up to this test: GMail and Basecamp. No wonder I use them both all the time!
There are clear advantages to this kind of design: it's typically lighter on bandwidth (using fewer graphics), has a focus on content (especially if it's text), and allows accessibility for everyone.
Apple-Plus, people. Do it. Your eyes will thank you!
